Abstract:

In vitro sperm activation by adding some materials was found to be very important methods to deals with DNA fragmentation of sperms which was one of causes of male infertility. The objective of the present study was to investigate the effect of different concentrations of tamoxifen citrate(TC) on the outcome of in vitro human sperm activation. Onehundred semen samples with range of age (32.21± 0.83) years and their mean duration of infertility (5.26 ± 0.41) years.Before using centrifugation swim-up technique, the washed samples were divided into 3 groups: control group (G1) without tamoxifen citrate. While in G2 and G3,two concentrations of tamoxifen citrate (20µg/ concentration of TC when compared with other treated group. The results of DNA fragmentation in post-activation revealed a significant decreased mL and 40µg/mL) respectively were post-activation enriched with SMART medium.Then, sperm DNA fragmentation was evaluated after the addition (P≤0.05) in treated groups (G2 and G3), compared with pre-activation and control group (G1).Based on the results of this study, it can be concluded that the addition of 40µg/mL of TC to washed sperms can improve sperm DNA integrity in vitro.oftamoxifen citrate.
