Two hundred and ten specimens include urine ,blood and ear swab were collected from different hospitals in Baghdad city; 85 (40%) isolates were diagnosed as Proteus spp. Proteus isolates were recorded (82%) , (11.7%) and (5.8%)in urine, blood and ear swab samples respectively. PCR technique was shown 30(35.3%)isolates positive for Urease C gene that specific for Proteus vulgaris.The ability for chondritinase production was investigated, 24(80%)isolates positive for chondritinase production but only one isolate have higher enzyme activity at (175.2U/ml).
The Chondroitinase was purified by short steps included precipitate with 60% ammonium sulfate , dialysis and gel filtration by Sephadex G-150; the enzymatic activity was peaked to (270U/ml)andspecific activity (587U/mg)with 23.6% yield. The enzymatic activity for purified enzyme was increased significantly to (563.7U/ml) after treatment with 1M of tyrosine and reached to (633U/ml) after treatment with 50% of vitamin k,and reached to (610.3U/ml)after treatment with 50% of piroxicam which know(Feldene).The aim of this study investigate effect of amino acids, vitamins and drugs with different concentrations on enzyme activity.
